What Is Selank?

Selank is a synthetic heptapeptide — Thr-Lys-Pro-Arg-Pro-Gly-Pro. Molecular weight 751.87.

Its parent molecule is tuftsin — and tuftsin’s origin is the interesting part.

Tuftsin comes from an antibody

Tuftsin is a naturally occurring tetrapeptide: Thr-Lys-Pro-Arg. It is not synthesised as a peptide in its own right. It is cleaved out of the Fc region of immunoglobulin G — the constant tail of an antibody.

So the body produces tuftsin by cutting up an antibody. Its known function is immune: it modulates macrophage activity and phagocytosis.

Selank is that immune peptide, extended and stabilised — and it turned out to have effects on anxiety that the parent molecule does not have. That’s the surprise: an antibody fragment that acts on the brain.

The stabilising tail

Researchers at the Institute of Molecular Genetics added a Pro-Gly-Pro C-terminal extension to protect tuftsin from enzymatic degradation.

The same tail appears on Semax — built at the same institute from ACTH, a pituitary hormone fragment. Same engineering solution, entirely different parent, entirely different effect.

How Selank Works: Anxiolysis Without the Benzodiazepine Mechanism

This is the point of the compound.

Benzodiazepines work by binding the GABA-A receptor directly at the benzodiazepine site. That is effective — and it is why they sedate, why tolerance develops, and why dependence follows.

Selank is described in the literature as a positive allosteric modulator of GABA that does not bind the benzodiazepine site. The reported profile is anxiolytic activity without sedation and without dependence liability — and that combination is the reason it was pursued at all.

Research also reports:

  • Serotonergic modulation — effects on serotonin metabolism
  • BDNF expression — overlapping with, but weaker than, Semax
  • Immunomodulatory activity — inherited from the tuftsin lineage. Effects on cytokine expression and macrophage activation.
  • Enkephalin metabolism — effects on endogenous opioid peptide turnover

The neuro-immune combination is genuinely unusual. Selank is one of very few compounds acting on anxiety and on immune signalling through the same molecule — a direct consequence of having been built from an antibody fragment.

Selank Benefits: What the Research Reports

Anxiolytic activity

The central finding, characterised by Seredenin and colleagues (1998). Rodent behavioural models report anxiolytic effects without the sedation associated with benzodiazepines. This is the basis of its registration as a medicine in Russia.

Neuro-immune effects

Cytokine expression, macrophage activation, interferon-related activity — inherited from tuftsin. Absent from Semax’s profile entirely.

BDNF and monoamines

Reported effects on BDNF expression and on serotonin metabolism, overlapping with Semax but less pronounced.

Enkephalins

Effects on endogenous opioid peptide metabolism.

Selank Dosage in the Published Research

Animal models

Rodent work reports microgram-to-milligram per kilogram quantities, most commonly administered intranasally — the same olfactory route used for Semax, and for the same reason: peptides do not readily cross the blood–brain barrier.

Humans

Human clinical work exists in Russia, where Selank is a registered medicine. That literature is largely Russian-language and has seen limited independent Western replication. No dose has been established as safe or effective by any regulatory authority outside Russia.

CompoundPlug provides no dosing recommendations.

Selank Side Effects Reported in Studies

Reported adverse findings are limited. The literature describes a favourable profile — specifically, an absence of the sedation, tolerance, and dependence associated with benzodiazepine anxiolytics.

The evidential caveat is the same as for Semax: a favourable record from a body of research with limited independent Western scrutiny is weaker evidence than the same record fully replicated. That is a statement about the evidence base, not the compound.

Selank Research Applications

  • Anxiolytic mechanism — GABAergic modulation without benzodiazepine-site binding
  • Neuro-immune interface — cytokine expression, macrophage activation; the tuftsin lineage
  • Tuftsin analog pharmacology — structure–activity; what the Pro-Gly-Pro extension adds
  • BDNF expression — comparative with Semax
  • Serotonergic modulation — serotonin metabolism
  • Enkephalin metabolism — endogenous opioid turnover
  • Intranasal delivery — olfactory route to the CNS

All applications preclinical, in-vitro / in-vivo only.

Selank is not an authorized health product in Canada. No DIN, no NPN, not reviewed by Health Canada, not approved for human use.

It is not a controlled substance under the CDSA.

Note: Selank is a registered medicine in Russia. That has no bearing on Canadian status.

CompoundPlug supplies Selank strictly as a research chemical. See Research Use Only.

Selank Reconstitution & Storage

Water-soluble. Bacteriostatic water, added slowly down the vial wall. Swirl, never vortex.

Storage: lyophilized and sealed, up to 24 months. Reconstituted, 2–8 °C, 3–4 weeks, protected from light.

Quality & Verification

Independent HPLC and mass-spec testing, published rather than asserted.

Same small-peptide problem as Semax. At seven residues, Selank is quick and cheap to synthesise — and easy to produce with a deletion sequence, a six-residue impurity missing one amino acid.

Mass spectrometry confirms the molecular weight is 751.87, matching the full sequence. HPLC quantifies how much of the vial is intact compound. On a heptapeptide, those two tests are the whole assurance.

Vials measured by peptide content. Published analysis in the COA Library.

Frequently Asked Questions

What is Selank?

A synthetic heptapeptide — Thr-Lys-Pro-Arg-Pro-Gly-Pro, MW 751.87 — built from tuftsin, with a Pro-Gly-Pro stabilising tail. Developed at the Institute of Molecular Genetics, Russian Academy of Sciences.

What is tuftsin?

A naturally occurring tetrapeptide (Thr-Lys-Pro-Arg) that the body produces by cleaving it out of the Fc region of an antibody — immunoglobulin G. Its known function is immune: it modulates macrophage activity and phagocytosis. Selank is that immune peptide, extended and stabilised — and it turned out to have anxiolytic effects the parent does not.

How does Selank work?

It is described as a positive allosteric modulator of GABA that does not bind the benzodiazepine site. The reported profile is anxiolytic activity without the sedation, tolerance, and dependence liability associated with benzodiazepines. It also carries immunomodulatory activity inherited from tuftsin — a neuro-immune combination that is genuinely unusual.

What are the reported benefits of Selank?

Anxiolytic effects in rodent behavioural models without sedation (Seredenin et al., 1998) — the basis of its registration in Russia. Also reported: cytokine and macrophage effects from the tuftsin lineage, BDNF expression, serotonergic modulation, and effects on enkephalin metabolism.

Selank vs Semax — what’s the difference?

Same institute, same Pro-Gly-Pro stabilising tail, opposite parent molecules. Selank is built from tuftsin, an immune peptide cleaved from an antibody, and works through GABAergic and immunomodulatory pathways — the anxiolytic one. Semax is built from ACTH(4-7), a pituitary hormone fragment, and works through BDNF and NGF upregulation — the cognitive one. Non-overlapping mechanisms.

What is the Selank dosage in research?

Rodent studies report microgram-to-milligram per kilogram quantities, most commonly intranasal. Human clinical work exists in Russia. CompoundPlug provides no dosing recommendations.

Is Selank legal in Canada?

Not an authorized health product in Canada — no DIN, no NPN, not approved for human use. Not a controlled substance. It is a registered medicine in Russia, which has no bearing on Canadian status.

How is Selank reconstituted?

Water-soluble. Bacteriostatic water, added slowly down the vial wall, swirled — never shaken or vortexed. Molar calculations use MW 751.87.
