What Is Melanotan 2?
Melanotan 2 — written MT-2 or MT2, and both refer to the same compound — is a synthetic cyclic heptapeptide, an analog of α-MSH (alpha-melanocyte stimulating hormone), the endogenous peptide that signals through the melanocortin receptor system.
Its defining property is that it is non-selective
MT-2 binds all four principal melanocortin receptors: MC1R, MC3R, MC4R, and MC5R. The research literature attributes its broad and somewhat unpredictable effect profile directly to that lack of selectivity.
These receptors do entirely different things:
- MC1R — melanogenesis. Melanin synthesis in melanocytes.
- MC3R / MC4R — central signalling. Appetite regulation, and the sexual-function pathways in the hypothalamus.
- MC5R — exocrine gland function.
A single compound activating all of them will produce effects across all of them. That is the mechanistic reason MT-2’s effect profile is as broad as it is, and it is the reason a more selective molecule was eventually engineered out of it.
Melanotan 2 and PT-141: The Compound That Came Out of It
This is the most consequential thing in MT-2’s history, and it is worth understanding before anything else.
During melanocortin research, effects on sexual-function pathways were observed as an unintended finding — a consequence of MC4R agonism, in studies whose endpoint was something else entirely.
That observation did not stay a footnote. Palatin Technologies engineered a molecule to isolate it: keep the MC3R and MC4R activity, drop the MC1R activity responsible for melanogenesis.
The result was bremelanotide — PT-141 — which went on to become the first melanocortin receptor agonist ever approved by the FDA (2019).
MT-2 is the non-selective parent. PT-141 is what happens when you remove the receptor you didn’t want. The two compounds are a case study in why selectivity is a design goal rather than a technicality.
Melanotan 2: What the Research Reports
Melanogenesis
Increased melanin synthesis via MC1R agonism. This is the most-studied effect and the one the compound was originally developed to investigate. Melanocyte models report increased melanin production.
Appetite and food intake
MC4R agonism affects appetite regulation, and rodent models report effects on food intake. MC4R is one of the better-characterised appetite receptors in the hypothalamus.
Sexual-function pathways
Also MC4R-mediated. This is the observation that led directly to the development of PT-141.
Photoprotection research
UV-response models — the original research context, given melanin’s role in UV absorption.
All findings are preclinical or from early-stage work. MT-2 has never completed clinical development in any jurisdiction.
Melanotan 2 Dosage in the Published Research
The MT-2 literature is limited and largely preclinical — and that is itself the most important thing to say about it. Unlike PT-141, which went through Phase 3, MT-2 was never carried through development.
Preclinical
Studies report quantities in the microgram-per-kilogram range, administered subcutaneously.
Early human work
Early melanocortin research reported quantities in the low-milligram range per administration. That work was small and was not carried through to development — and the reason it stopped is relevant to any assessment of the compound.
No dose of MT-2 has been established as safe or effective by any regulatory authority. CompoundPlug provides no dosing recommendations.
Melanotan 2 Side Effects Reported in Studies
MT-2 has a more substantial adverse-effect record than most compounds in this catalog, and it is a direct consequence of the non-selectivity.
Commonly reported
- Nausea — attributed to central melanocortin activation
- Flushing
- Spontaneous erections — MC4R agonism, the effect PT-141 was built to isolate
- Darkening of existing nevi and freckles — MC1R agonism acting where melanocytes already are
⚠️ The dermatological concern
This must be stated plainly, and it is specific to melanogenic compounds rather than a generic caution.
Case reports in the medical literature describe changes in the appearance of existing nevi — moles — following MT-2 use, and have raised questions about melanoma risk.
The mechanistic concern is coherent rather than speculative: MT-2 stimulates melanocytes. Melanoma is a cancer of melanocytes. A compound that drives melanocyte activity, applied to a person who already has atypical melanocytes, raises an obvious and unresolved question.
This is an active concern in the literature, not a theoretical one. It is the primary reason MT-2 is studied in cell culture rather than administered, and it is a reason regulators have taken an interest.
Regulatory
MT-2 was placed on the FDA’s Category 2 restricted list for pharmacy compounding in 2023; the restriction was lifted in 2026 without addition to the permitted list. Health Canada has issued warnings regarding melanotan products sold in Canada.
MT2 Peptide, MT-2, Melanotan II: The Naming
MT2 peptide, MT-2, MT II and Melanotan II all refer to this compound. The variation is orthographic rather than chemical — the hyphen and the roman numeral are conventions, not distinctions, and no supplier or paper using one form is describing a different molecule from one using another.
Two naming distinctions that are real and worth keeping straight:
- Melanotan 1 is a different compound. Afamelanotide — also called melanotan-1 or MT-1 — is a linear α-MSH analog with far greater MC1R selectivity. It is not stocked here, and it is not interchangeable with MT-2 in any research context.
- “Melanotan” without a number is ambiguous and appears in the literature and in regulatory notices referring to either. Where a source does not specify, the compound is not identified.
This page, and the vial it describes, is Melanotan 2 — the non-selective heptapeptide, molecular weight approximately 1024.2.

Quality & Verification
Independent HPLC and mass-spec testing, published rather than asserted.
MT-2 and PT-141 are structurally close — and that is exactly why identity testing matters on this compound.
Both are cyclic heptapeptides derived from α-MSH. Their molecular weights are approximately 1024.2 and 1025.2 — roughly one dalton apart.
They are pharmacologically different molecules. MT-2 is non-selective and activates MC1R; PT-141 has substantially reduced MC1R activity. That is the difference between a compound that drives melanogenesis and one that does not — and a purity test cannot see it.
Mass spectrometry resolves compounds a single dalton apart. On these two, it is the only test that identifies which is in the vial.
Vials measured by peptide content. Published analysis in the COA Library, tied to the lot.

Frequently Asked Questions
What is Melanotan 2?
A synthetic cyclic heptapeptide analog of α-MSH, and a non-selective melanocortin agonist — it binds MC1R, MC3R, MC4R, and MC5R. That non-selectivity is its defining property and the source of its broad effect profile.
Is MT2 peptide the same as Melanotan 2?
Yes. MT2 peptide, MT-2, MT II and Melanotan II are all the same compound — the variation is spelling, not chemistry. Melanotan 1 is a different molecule: afamelanotide, a linear α-MSH analog with much greater MC1R selectivity, and not interchangeable with MT-2. “Melanotan” with no number is ambiguous and should not be treated as identifying either one.
What’s the difference between Melanotan 2 and PT-141?
PT-141 is the refined version. During MT-2 research, effects on sexual-function pathways were observed as an unintended MC4R finding. Palatin engineered a molecule to isolate it — keeping MC3R/MC4R activity and dropping the MC1R activity that drives melanogenesis. That molecule, bremelanotide, became the first FDA-approved melanocortin agonist. MT-2 is the non-selective parent.
What effects does the research report?
Increased melanin synthesis via MC1R. Effects on appetite and food intake via MC4R in rodent models. Effects on sexual-function pathways, also MC4R-mediated — the observation that led to PT-141. All preclinical; MT-2 never completed clinical development in any jurisdiction.
What are the side effects of Melanotan 2?
Nausea, flushing, spontaneous erections, and darkening of existing nevi and freckles — all consequences of the non-selectivity. More seriously: case reports describe changes in the appearance of existing moles and have raised questions about melanoma risk. The mechanistic concern is coherent — MT-2 stimulates melanocytes, and melanoma is a cancer of melanocytes. This is an active concern in the literature.
Is Melanotan 2 legal in Canada?
Not an authorized health product in Canada — no DIN, no NPN, not approved for human use. Health Canada has issued public warnings about unauthorized melanotan products. It is not a controlled substance. Supplied research-use-only.
How is Melanotan 2 reconstituted?
Water-soluble. Bacteriostatic water, added slowly down the vial wall, swirled — never shaken or vortexed. See the Melanotan 2 calculator.
